Traceability of Sicilian Durum Wheat Landraces and Historical Varieties by High Molecular Weight Glutenins Footprint

چکیده

Over the last new decade, there has been a strong interest in landraces and historical wheat varieties from farmers, manufacturers consumers. They are agronomically nutritionally interesting but supply chain (from seed to end-product) is not solid traceable. High molecular weight glutenins (HMW-GS) can act as markers trace varietal correspondence verify genetic purity of grain consequently flours, marketed labeled mono-varietal. In present work, HMW-GS different durum Sicilian (Timilia, Russello, Perciasacchi) one variety Margherito were analyzed. At first, specific protein profiles assigned each landrace by SDS-PAGE MALDI-TOF/MS analyses, thanks availability pure seeds. Analysis then carried out random samples batches same grown on farm South-East Sicily. The results highlighted presence bands within individual batches, which reflected complex corresponding commercial flours labelled bread Maiorca cultivated was also found contaminant at percentages batches. this study offer opportunities improve or cultivated, underlining need for accurate controls field transformation process be mono-varietal products.

Phenotypic Diversity of Tunisian Durum Wheat Landraces

Tunisia is considered as a diversification centre of durum wheat (Triticum durum Desf.) and barely (Hordeum vulgare). It is characterised by large genetic diversity; however, several genotypes are in the process of disappearance. The safeguard and rehabilitatation of this genetic inheritance requires genetic characterisation, evaluation and in-situ and ex-situ conservation. In this context, a l...

High‐density molecular characterization and association mapping in Ethiopian durum wheat landraces reveals high diversity and potential for wheat breeding

Durum wheat (Triticum turgidum subsp. durum) is a key crop worldwide, and yet, its improvement and adaptation to emerging environmental threats is made difficult by the limited amount of allelic variation included in its elite pool. New allelic diversity may provide novel loci to international crop breeding through quantitative trait loci (QTL) mapping in unexplored material. Here, we report th...

Aromatic and proteomic analyses corroborate the distinction between Mediterranean landraces and modern varieties of durum wheat

In this paper volatile organic compounds (VOCs) from durum wheat cultivars and landraces were analyzed using PTR-TOF-MS. The aim was to characterize the VOC's profile of the wholemeal flour and of the kernel to find out if any VOCs were specific to varieties and sample matrices. The VOC data is accompanied by SDS-PAGE analyses of the storage proteins (gliadins and glutenins). Statistical analys...

Separation of durum wheat proteins by ultrathin-layer isoelectric focusing: a new tool for the characterization and quantification of low molecular weight glutenins.

An isoelectric focusing method capable of resolving all groups of storage protein of the wheat seed, including the most basic low molecular weight glutenin (LMWG), was developed. Ultrathin polyacrylamide gels were used after drying and rehydration with 8 M urea, 50 mM DTE and 2.4% carrier ampholytes (pH 4-9). Densitometric scanning of the isoelectric focusing gels permitted a more accurate and ...

Spatial and Temporal Genetic Structure of Wild Emmer Wheat in Jordan. Ii. High-molecular-weight Glutenins and Allozymes

A study was designed to sample and characterize 12 populations of wild emmer wheat in Jordan at the high-molecular-weight (HMW) glutenin and allozyme levels. Data were collected and individual plants were sampled from 207 permanent sampling points in three central, three peripheral, and six marginal populations during all or part of a five-year study period. Patterns of variation, based on four...
